Acclimatizing Tissue Culture Plants Before They Become Mother Stock

Tissue culture plants that have just been removed from the bottle are not yet “field-ready seedlings” or “mother plants ready for use.” While growing inside the culture vessel, they have been kept under sterile conditions, high humidity, controlled light, and a constant supply of nutrients from the agar medium. Once they are transferred to a greenhouse, the plantlets must shift into an entirely new way of living. They must move from depending on nutrients inside the bottle to photosynthesizing on their own, absorbing water through their roots, regulating transpiration, and adapting to real environmental conditions. Acclimatization is therefore a critical transition stage that directly affects the quality of future planting material.

Tissue culture plantlets are delicate after deflasking because their leaves, roots, and physiological systems are not yet as developed as plants grown outside the bottle. Leaves formed in vitro often have an underdeveloped cuticle, less functional stomata, and are accustomed to very high humidity. When these plantlets are moved into drier air, water can evaporate from the leaves quickly, causing wilting or transplant shock. For example, newly deflasked banana or philodendron plantlets placed immediately in strong wind or direct sunlight may droop, wilt, and stop growing within only a few hours.
The root system is another point that requires careful attention. Although plantlets inside the bottle may already have roots, those roots developed in agar medium, not in real growing substrate. As a result, they may not yet absorb water and nutrients efficiently outside the vessel. After transplanting, some of the original roots may be damaged or slow to adapt, and the plant must produce new roots suited to greenhouse conditions. For example, a plantlet that looks beautiful inside the bottle but has very few or very short roots may be harder to establish than a plantlet with a stronger root system and a more developed shoot.
The first step in acclimatization is selecting suitable plantlets before deflasking. Only plantlets with healthy leaves, shoots, and roots should be chosen. They should not be yellow, bruised, contaminated, or abnormal, and their size should be relatively uniform. Plantlets that are too small, poorly rooted, or visibly abnormal should be separated and held back rather than mixed with the main batch, as they can increase losses in the greenhouse. After removing the plantlets from the bottle, agar must be washed carefully from the roots and crown. Agar contains sugar and residual nutrients, and if it remains on the plant, it can become an easy food source for fungi and bacteria in the growing medium.
The first acclimatization stage, often called primary hardening, is the most important period. During this stage, the plantlets are adapting from the protected in vitro environment to real greenhouse conditions. They should be kept under low to moderate light, with high humidity, but not in a stagnant environment that encourages fungal growth. The growing medium must be clean, airy, well-drained, and able to hold enough moisture without becoming waterlogged. The key watering principle is “moist but not wet.” If the medium stays too wet, young roots may be deprived of oxygen and become vulnerable to root rot. For example, tissue-cultured banana plantlets may be started in cocopeat or another airy medium that holds moisture well, then gradually adjusted once new leaves and roots begin to form.
Once the plantlets begin to establish, they enter the secondary hardening stage. The goal at this point is to strengthen the plants gradually by reducing humidity, increasing light, improving air movement, and encouraging greater dependence on photosynthesis. At this stage, the plant should begin producing new leaves formed outside the bottle and new roots that function properly in the growing medium. For foliage plants such as Monstera, Philodendron, or Aglaonema, the appearance of new leaves in the greenhouse is a good sign that the plant is adapting successfully and may soon be ready for transfer to a larger pot or grow bag.
The choice of growing medium has a major effect on survival and plant development. A suitable medium should be clean, airy, well-drained, and not overly water-retentive. Common options include cocopeat mixed with perlite, peat moss, vermiculite, carbonized rice husk, or other porous materials depending on the plant species. Bananas may require a medium that retains moisture well but does not become compacted. Variegated foliage plants should be grown in clean, well-draining media to reduce stem and crown rot. Bulbous or rhizomatous plants require extra caution because young crowns and storage organs can rot easily after deflasking if water accumulates around them.
When the goal is to produce mature mother stock, acclimatization should not be evaluated only by survival. Plant quality must be selected carefully. A plant suitable for mother stock should produce new leaves continuously, have a strong stem, develop white and well-branched roots, show no disease or insect problems, display no virus-like symptoms, and remain true to type. Plants with abnormal leaves, off-color foliage, slow growth, dwarfing, or off-type characteristics should be removed from the mother stock group immediately. For example, a variegated plant with a pattern that differs significantly from the selected mother line, or a banana with abnormal plant form, should not be used as stock material for further production.

For commercial production, the acclimatization greenhouse should be managed as a semi-clean production area. It does not need to be sterile like a laboratory, but it must be clean and well organized. Production batches should be labeled, varieties should be separated, deflasking dates should be recorded, insect vectors should be controlled, sick plants should be removed promptly, and water splash from diseased plants to healthy plants should be avoided. Beneficial microorganisms such as Trichoderma, Bacillus, or arbuscular mycorrhizal fungi may support root development and disease suppression in some systems, but they should be selected according to the plant species and tested first, as results depend on the growing medium, greenhouse conditions, and plant variety.
In summary, acclimatizing tissue culture plants before they become mature mother plants is the process of transforming delicate in vitro plantlets into strong planting material that can be used for further propagation. It is not simply about keeping the plants alive. The goal is to produce plants with strong roots, healthy new leaves, sturdy stems, disease-free growth, true-to-type characteristics, and enough uniformity to serve as reliable mother stock.
If poor-quality plants are selected as mother stock, abnormalities or diseases may be multiplied on a large scale in future production. Proper acclimatization is therefore the essential bridge between laboratory work and commercial planting material production.
